Basic Information about Vardenafil
Basically, this medication comes as pills in 120 mg doses, and two main ingredients that are present in any pill are vardenafil and sildenafil in a 1:5 ratio. This means that every Silvitra tablet contains 20 mg of vardenafil and 100 mg of sildenafil.

It plays an important role in erectile dysfunction treatment, though doesn’t provide protection against such STDs as HIV, syphilis, hepatitis, etc.
Vardenafil in Silvitra is considered to be a safe and well-tolerated compound in patients with ED. It is monohydrochloride salt and a potent selective inhibitor of cGMP (guanosine monophosphate) specific PDE-5 (phosphodiesterase type). It has a quite solid texture, is almost colorless and soluble in such compounds as ethanol, methanol and acetone.
Mechanism of Action
Vardenafil increases tumescence and rigidity of the penis with the help of a similar mechanism of action as in other PDE-5 inhibitors. It provokes an erection inhibiting cGMP PDE-5 by sending a signal to the corpus cavernosum, the penile tissue responsible for erection occurrence. This results in endogenous increase of cGMP levels in the erectile tissue of the penis and its vessels, and that is why dilatation of corporeal sinusoids increase, which makes it possible for more blood to flow in, thus, inducing erection. However, it’s important to note that erection occurs only in case nitric oxide is released from endothelial cells in the penile tissue and neurons, which is possible only during sexual stimulation. Nitric oxide activates guanylyl cyclase, the enzyme that helps convert guanosine triphosphate into cGMP. This process of conversion results in activation of protein kinase, diminishing of calcium levels in the cells, phosphorylation of a few proteins, smooth and slow smooth muscle relaxation and increased flow of the blood resulting in corpus cavernosum enlargement. As a result of increased tumescence the veins between tunica albuginea and corpus cavernosum compress, and this provokes a stiff erection. Therefore, vardenafil is only effective if there is sexual arousal.
